Cover Illustration: Superposition of the structures of two unrelated fluorescein-binding antibodies, FITC-E2 (PDB 2A9N) and 4-4-20 (PDB 1FLR), shows that the two antibodies bind the hapten in different orientations. Fluorescein-binding antibodies were used as a model system to study the interactions between antibody and hapten and to analyze the correlation between macroscopic binding affinities, association and dissociation rates with the single molecule rupture forces measured by atomic force microscopy and with the unbinding pathways observed in molecular dynamics simulations of the forced unbinding experiments.
